About Flow Cytometry

Flow cytometry is a technique for counting, examining and sorting microscopic particles suspended in a stream of fluid. It allows simultaneous multiparametric analysis of the physical and/or chemical characteristics of single cells flowing through an optical and/or electronic detection apparatus. By combining this with fluorescent tagging, vast amounts of data can be collected.

 

Training Information

The CIC staff will provide training for interested individuals wanting to learn more about flow cytometry and how to operate the flow analyzers. Training can be scheduled by e-mailing Daniel Jackson at JacksonDan@health.missouri.edu. Training sessions are mandatory and are offered on Tuesday and/or Thursday afternoons from 3-5 pm at a cost of $40.00 per training in addition to the fee for using the instrument during the training. If this time does not work, please let the CIC know and we may be able to accommodate to your availability. The official training consists of two parts. The first being a theoretical lesson of flow cytometry and the policies of the CIC facility. The second part involves time on the instrument and a guided run through of a practice experiment. After this is completed, we may require you to run with a CIC operator until you feel comfortable with the instrument and software.

 

The MoFlo XDP has set the standard for cell sorting with both functionality and speed. It is designed for researchers with features of improved productivity, an array of bio-safety features and easy-to-use software. It is capable of up to 9 parameter/7 color sorting or analysis. The high-speed and configurable system has solid state lasers, high viability, high yield, an analysis rate of 100,000 events per second, and sort rates of up to 70,000 events per second. This allows the user to obtain high numbers of rare populations in a greatly reduced amount of time!

The BD LSRFortessa X-20 is configured with 4 lasers (488 nm, 640 nm, 405 nm, and 552 nm) capable of detecting of up to 15 colors and 17 parameters. The dual forward scatter detection system includes a highly sensitive forward scatter PMT, allowing for the recognition of particles as small as 0.2 micron. In addition, the multi-well plate loader offers the flexibility to process samples directly from multi-well plates, making it amenable to high throughput applications. The instrument is equipped with the FACSDiva 8.0 Software and is available 24/7 after appropriate training.

The CyAn ADP High-Performance Flow Cytometer is a state-of-the-art flow cytometer that utilizes multiple laser excitation sources to analyze biological cells, beads, or other microscopic particles. The CyAn ADP has state-of-the-art optics, utilizes high performance digitization with pulse processing speed up to 70,000 events per second complementing a high acquisition and analysis capability of up to 100,000,000 events. Up to 9 x 9 channel compensation can be performed manually, using bi-exponential transformation (VisiComp) or automatically.

The FACScan is a highly sensitive nonsorting instrument equipped with a 488nm-argon laser that is used exclusively for quantitative analysis applications. Since the 'Scan is equipped with three fluorescence detectors, simultaneous analysis of five parameters (two light scatter and three fluorochromes) is routine. FITC, PE, PI, and PerCP are the dyes most commonly used, but new dyes are continually being developed, expanding the range of this system. The FACScan is engineered with fixed optical, electronic and fluidic components, giving it the flexibility to function as an investigator-operated instrument after training.

The FACSCalibur is equipped with dual excitation lasers at the 488nm and 633nm wavelengths for quantitative analysis. The 'Calibur is also equipped with a sorting option for fragile cells unsuited for the stresses of high speed sorting. Since the 'Calibur is equipped with four fluorescence detectors, simultaneous analysis of six parameters (two light scatter and four fluorochromes) is routine. Possessing fixed optical, electrical, and fluidic components, this system can also be utilized without operator assistance after training.
